Detection Principle
考马斯亮蓝G-250染料游离状态下呈棕红色,与蛋白质结合后变为蓝色,染料的最大吸收峰由465 nm变为595 nm,在一定浓度范围内蛋白质-染料复合物吸光值与蛋白质含量成正比,通过测定595 nm处吸光值的变化即可定量检测蛋白质的含量。

Notices
① Bradford法测定蛋白浓度不受绝大部分样品中的化学物质的影响,样品中巯基乙醇的浓度可高达1 M,二硫苏糖醇的浓度可高达5 mM;但受略高浓度的去垢剂影响,需确保样品中SDS浓度低于0.1%,TritonX-100低于0.1%,Tween 20、60、80低于0.06%;
