Detection Principle
叶绿素a和叶绿素b分别在645 nm和663 nm处具有特征吸收峰,根据经验公式即可计算叶绿素a和叶绿素b以及总叶绿素的含量。

  • 检测波长: 645 nm & 663 nm
  • 信号响应: 递增型
Notices

①叶绿素对光敏感,研磨和提取等操作尽量避光或者在弱光下进行;

②提取时应浸提至组织残渣完全变白,否则会因提取不充分而影响测定结果;

③提取液冲洗研钵时应冲洗至所有的绿色物质转移至试管/离心管中;

④若测定吸光值超过1.0,建议将浸提液使用提取液适当稀释后再进行测定,计算时相应调整(按实际稀释倍数调整计算公式中D);若测定吸光值小于0.05时,可适当减少提取过程中提取液的用量后再进行测定,计算时相应调整(按实际提取后总体积调整计算公式中V提);

注: 为保证结果准确且避免试剂损失,测定前请仔细阅读说明书(以实际收到说明书为准),确认试剂储存和准备是否充分,操作步骤是否清楚,且务必取2-3个预期差异交的样本进行预测定,过程中问题请您及时与工作人员联系。
