Detection Principle
丙酮酸脱氢酶能够催化丙酮酸脱羧生成羟乙基-TPP,能够将2,6-二氯酚靛酚(2,6-DCPIP)还原,2,6-二氯酚靛酚在605 nm处有特征吸收峰,通过吸光值的变化即可表征丙酮酸脱氢酶活性。

Notices

①测定过程中所有样本在冰上放置,以免变性和失活;

②准确在60 s处完成读数,以保证实验结果的准确性,若使用96孔板应使用多道移液器并分批进行检测;

③∆A测定应在0.01-0.25之间,若∆A测定大于0.25,需将样本稀释后再进行测定,计算时相应修改;

④提取液中含有一定浓度的蛋白(约1 mg/mL),测定样品蛋白浓度时需要减去提取液自身的蛋白含量;

注: 为保证结果准确且避免试剂损失,测定前请仔细阅读说明书(以实际收到说明书为准),确认试剂储存和准备是否充分,操作步骤是否清楚,且务必取2-3个预期差异交的样本进行预测定,过程中问题请您及时与工作人员联系。
