Detection Principle
丙酮酸脱氢酶能够催化丙酮酸脱羧生成羟乙基-TPP，能够将2,6-二氯酚靛酚（2,6-DCPIP）还原，2,6-二氯酚靛酚在605 nm处有特征吸收峰，通过吸光值的变化即可表征丙酮酸脱氢酶活性。

Notices
①测定过程中所有样本在冰上放置，以免变性和失活；
②准确在60 s处完成读数，以保证实验结果的准确性，若使用96孔板应使用多道移液器并分批进行检测；
③∆A测定应在0.01-0.25之间，若∆A测定大于0.25，需将样本稀释后再进行测定，计算时相应修改；
④提取液中含有一定浓度的蛋白（约1 mg/mL），测定样品蛋白浓度时需要减去提取液自身的蛋白含量；
注： 为保证结果准确且避免试剂损失，测定前请仔细阅读说明书（以实际收到说明书为准），确认试剂储存和准备是否充分，操作步骤是否清楚，且务必取2-3个预期差异交的样本进行预测定，过程中问题请您及时与工作人员联系。
