Detection Principle
羧酸酯酶能够催化乙酸-1-萘酯生成萘酯，萘酯进一步与固蓝反应生成偶氮化合物，产物在450 nm处具有特征吸收峰，通过吸光值变化即可表征羧酸酯酶的活性。

Notices
①试剂二配制后室温条件下稳定性较差，建议配制和使用过程中始终保持冰上放置，若试剂变色则停止使用；建议测定过程中根据使用量取用试剂二置于冰上放置，剩余试剂应及时置于-20℃保存；
②试剂二和试剂四配制后有效期较短，为便于试验安排均附赠一瓶作为备用；
③准确在10 s和310 s处完成吸光值测定，以保证实验结果的准确性和重复性；同时测定多个样本时，应使用多道移液器且分批进行测定，以确保组间反应时间一致；
④若A2测定或∆A大于1.0，建议将粗酶液使用提取液适当稀释后再进行测定；若∆A小于0.02，建议制备更高浓度样本、增加上样量（粗酶液增加至20 μL，其它试剂量不变）或适当延长酶促反应时间（37℃反应时间延长至10-30 min）后再进行测定，计算时相应修改；
注： 为保证结果准确且避免试剂损失，测定前请仔细阅读说明书（以实际收到说明书为准），确认试剂储存和准备是否充分，操作步骤是否清楚，且务必取2-3个预期差异交的样本进行预测定，过程中问题请您及时与工作人员联系。
