Detection Principle
氨基酸中的α-氨基在加热条件及弱酸环境下能够与水合茚三酮反应生成蓝紫色化合物,产物在570 nm处具有特征吸收峰,通过吸光值变化即可定量检测氨基酸的含量。

Notices

①试剂二和试剂三均需现用现配且避光保存,配制后有效期较短,为便于试验安排,均附赠一瓶作为备用;

②若测定吸光值超出标准吸光值线性范围:高于最高值建议将待测样本使用试剂一适当稀释后再进行测定;低于最低值建议适当调整样本量和提取液比例后再进行测定,计算时相应调整;

③脯氨酸和羟脯氨酸与茚三酮反应在570 nm处无吸收峰,测定结果不含这两种氨基酸的含量;

④提取过程会使蛋白变性,若使用蛋白浓度计算,需单独使用PBS提取后再测定蛋白浓度;
