Detection Principle
待测样本经酸水解产生游离羟脯氨酸,被氯胺T氧化为吡咯,吡咯能够与对二甲氨基苯甲醛(DMAB)反应生成红色化合物,产物在560 nm处具有特征吸收峰,通过吸光值的变化即可定量检测羟脯氨酸的含量。

Notices

①若测定吸光值超出标准线性吸光值范围:高于最高值建议将待测样本适当稀释后再进行测定;低于最低值建议适当增加样本量后再进行测定,计算时相应修改;

②试剂具有一定的毒性,请操作时做好防护措施,防止吸入或与皮肤接触;

③测定蛋白浓度时需要使用PBS单独提取样本蛋白后进行测定;
