Detection Principle
H2O2/Fe2+通过Fenton反应产生羟自由基,将邻二氮菲-Fe2+水溶液中Fe2+氧化为Fe3+,导致536 nm处吸光值下降,根据吸光值下降速率的抑制程度即可表征样品清除羟自由基的能力。
  • 检测方法: Fenton比色法
  • 检测波长: 536 nm
  • 信号响应: 递增型
Notices

①不同样本羟自由基清除能力可能相差较大,若需要比较不同样本的羟自由基清除能力,建议对于同一批植物样本加入等量的样本,液体样本加入相同体积,提取物或者药物配制为相同浓度;将样本根据预实验结果进行适当调整,比较同样浓度(相同稀释倍数)的清除率大小;

②若测定样本较多时,可按试剂一:试剂二:试剂三=1:2:2的体积比配制为检测工作液,根据使用量现用现配;

③若A测定大于0.8,建议将待测样本使用提取液适当稀释后再进行测定;若A测定小于0.1,建议适当增加样本质量或液体样本体积重新提取后再进行测定,计算时相应修改;

注: 为保证结果准确且避免试剂损失,测定前请仔细阅读说明书(以实际收到说明书为准),确认试剂储存和准备是否充分,操作步骤是否清楚,且务必取2-3个预期差异交的样本进行预测定,过程中问题请您及时与工作人员联系。
