Detection Principle
谷胱甘肽和5,5’-二硫代-双-(2-硝基苯甲酸)(5,5’-dithiobis-2-nitrobenoic acid,DTNB)反应生成黄色的2-硝基-5-巯基苯甲酸和谷胱甘肽二硫化物(GSSG),2-硝基-5-巯基苯甲酸在412 nm处具有特征吸收峰,通过吸光值变化即可定量检测还原型谷胱甘肽的含量。

Notices

①样品处理过程应在冰上迅速完成,提取后待测样本应置于冰上保存且建议当天完成测定;

②试剂一中含有蛋白质沉淀剂,待测样本不能用于蛋白浓度测定,需另取组织使用1×PBS或生理盐水按照与待测样本相同的样本浓度重新提取后,再进行蛋白含量测定;

③若测定吸光值超出标准吸光值线性范围:高于最高值建议将待测样本使用试剂一适当稀释后再进行测定;低于最低值建议制备更高浓度样本后再进行测定,计算时相应修改;

注: 为保证结果准确且避免试剂损失,测定前请仔细阅读说明书(以实际收到说明书为准),确认试剂储存和准备是否充分,操作步骤是否清楚,且务必取2-3个预期差异交的样本进行预测定,过程中问题请您及时与工作人员联系。
