Detection Principle
ICDHc催化异柠檬酸生成α-酮戊二酸,并将NAD+还原为NADH,NADH在340 nm处具有特征吸收峰,通过吸光度的变化即可计算ICDHc的活性。
  • 检测方法: NADPH速率法
  • 检测波长: 340 nm
  • 信号响应: 递增型
Notices

①测定过程中试剂二、试剂三和样本需冰上放置,以免失活,工作液37℃水浴中放置;

②反应过程中应保持37℃恒温,建议在烧杯中装入适量37℃蒸馏水,将此烧杯放入37℃恒温水浴中,在反应过程中把比色皿连同反应液放在此烧杯中;

③准确在20 s和140 s处完成读数,以保证实验结果的准确性;

④若∆A测定和A1测定>0.5建议使用提取液将样本稀释后再进行测定,使∆A测定和A1测定<0.5以提高检测的灵敏度;

注: 为保证结果准确且避免试剂损失,测定前请仔细阅读说明书(以实际收到说明书为准),确认试剂储存和准备是否充分,操作步骤是否清楚,且务必取2-3个预期差异交的样本进行预测定,过程中问题请您及时与工作人员联系。
